I'm so sorry for the delay in getting back to you - it's taken us longer than we would like to respond!

Currently, there is no direct command-line support for creating a pull request with custom fields for GitHub Projects Beta (v2).

However, you could possibly do something like so:
gh pr create --project octocat-project

As for setting the status to "In Review", this is also not directly supported via the GitHub CLI. You would need to manually set your status on GitHub.

However, if your goal is to prevent the manual work of setting the status to "In Review" when a PR is created, I would recommend using the build-in Project workflow automations.
Using the built-in automations

For instance, if you are not currently utilizing the "Item added to project" workflow, you can use that and set it so that when a PR is added to the project to set its status to "In Review".
