Reactome Research Spotlight

[September 1, 2026] In their June 2026 Nature research article, "Cortical development dynamics across autism spectrum disorder mouse models", Schwarz et al. find that mutations linked to autism spectrum disorder (ASD) result in transient developmental delay of the brain cortex. A compendium of Reactome-derived protein-protein interactions (PPIs) was one of the resources used to construct a reference PPI network that was then used for network colocalization analysis of differentially expressed genes identified by single-nucleus multi-omic sequencing of brain samples from eleven monogenic mouse ASD models across three developmental stages. The authors show that ASD-linked mutations converge on perturbations of the radial glial cell lineage, with the largest transcriptional differences seen in neurons at early postnatal stages and involving downregulation of synaptic and ion channel-related genes.
